INNER-RETINAL COMPONENTS CONTRIBUTE TO THE A-WAVE OF THE ERG OF THE DARK-ADAPTED CAT. Robson J.G.1 and Frishman L.J.2 ’ Physiological Laboratory, Cambridge CE2 3EG UK/ 2 College of Optomern/ UniversitY of Houston, Houston. TX 77204 fUSA/ Purpose To determine the extent to which the initial negative wave and its return to baseline (the a-wave) of the flash electroretinogram IERG) can be modelled simply as the sum of a negative photoreceptor component and a positive bipolar-cell component. Method We have examined the time-course of the vitreally recorded a-wave in the urethane-anaesthetised cat and the effect upon it of suppressing the responses of inner-retinal Iamacrine or ganglion) cells either with intra-vitreal injection of n-methyl-oL-aspartate [NMoLA) or by application of a steady adapting background of moderate intensity known to affect mainly responses of inner retina. Resuhs The time-course of the a-wave at times more than about 10 msec after the flash is greatly altered by both NMDLA and adapting backgrounds. The most obvious effect of both procedures, removal of a negative component, occurred not only when the stimulus was weak enough for the negative STR to dominate under normal condidons, but also when the stimulus was strong enough for the rods themselves to contribute directly to the a-wave. However, once the responses of inner-retinal cells had been suppressed, the remaining a-wave could be satisfactorily explained as the sum of photoreceptor and bipolar-cell responses modelled as described by Lamb & Pugh I./. Physiol. 449, ~719, 1992) and Robson & Frishman (Vis. Neurosci. 12, 1995) respectively. Conclusion Inner-retinal components contribute substantially to the a-wave of the dark-adapted ERG. Only when these responses have been blocked pharmacologically, or have been sufficiently reduced by an adapting background, can the initial negative wave of the ERG be simply explained as the sum of photoreceptor and bipolar-cell components. Bradykinin Receptor Inhibition Affects the Rod b-wave in the Cat Electroretinogram
In 10 anaesthetized cats, electroretinographic (ERG) measurements were carried out to further elucidate the involvement of bradykinin as a substrate component of the renin-angiotensin system in retinal neurotransmission. Reducing angiotensin II concentration by angiotensin-converting enzyme (ACE) inhibition increased sensitivity (0.5 log units) and gain (50%) of the rod b-wave amplitude. The b-...
متن کاملPreparation of a Photoactivable Ligand for Bradykinin Receptors
ABSTRACT Most of physiological effects of bradykinin is due to its effect on bradykinin B2 receptors. There is no reports of preparation of a photoactivatable analogue of bradykinin reactive with sloubilized bradykinin B2 recpetors. Photoactivatable radioactive ligands are powerful tools in purification of receptors. In this study we report preparation of a photoactivatable iodinatable analo...
متن کاملRole of Bradykinin and Mas Receptor Blockade in Renal Vascular Responses to Angiotensin 1-7 in Adult Female Rats
Background and purpose: Chronic kidney disease is among the common diseases in the world. Studies show that women are more protected against renal diseases compared to men. On the other hand, vasodilatory axises of renin angiotensin system (angiotensin 1-7 (Ang 1-7)- Mas receptor (MasR)), kallikrein-kinin, and nitric oxide (NO) play key roles in kidney function and circulation in females. There...
متن کاملNoscapine antagonizes vasoconstrictor action of bradykinin in isolated human umbilical artery
Abstract Background: It has been demonstrated that noscapine, an antitussive opioid alkaloid, could antagonize bradykinin- induced responses such as bradykinin effects in guinea-pig ileum, cough induced by bradykinin receptor agonist and angiotensin converting enzyme inhibitors, and brain damage after brain edema both in neonatal rat model and in patients with stroke. In the present study, the...
متن کاملTorsional wave propagation in 1D and two dimensional functionally graded rod
In this study, torsional wave propagation is investigated in a rod that are made of one and two dimensional functionally graded material. Firstly, the governing equations of the wave propagation in the functionally graded cylinder derived in polar coordinate. Secondly, finite difference method is used to discretize the equations. The Von Neumann stability approach is used to obtain the time ste...
